It was whilst going through The Passover meal on Thursday, the night before Jesus would be crucified, with His disciples, that Jesus stopped at certain intervals to make a comment. These stops seem to coincide with the 4 cups that are part of a Passover Seder. After breaking bread and blessing the 3rd cup, Jesus spoke of it as The New Covenant in His blood. Jesus would not drink from the fruit of the vine again until He would drink it New with His disciples and all those at the Marriage Supper......
